文山野生金线莲的RAPD鉴别研究

摘要目的:采用RAPD技术,对金线莲DNA分子标记进行研究。方法:以文山地区药用金线莲原植物滇越金线兰及3种叶面特征不同的花叶开唇兰为材料提取基因组DNA,选择适合的随机引物和PCR反应体系,扩增分析金线莲的带谱。结果:16条引物共得到108条带谱、其中有95条多态性谱带,%,且4种材料都具特异性条带。结论:实验材料之间遗传差异性较大,花叶开唇兰可能已经形成了新的变型或变种;获得的特异性条带可用于区分干燥的金线莲药材。

Identification of 4 wild species of Anoectochilus from Wenshan by RAPD marker

Abstract: Objective: To analyze the DNA molecular characters of 4 samples of Anoectochilus with RAPD technology. Methods: With the genomic DNA as templates extracted from 4 samples of Anoectochilus. The RAPD PCR reaction systems and the random promers had been screend to amplify the molecular fragmentd of Anoectochilus. Results: 16 random primers produced 108 clear and stable number of polymorphic bands was 95, and the percentage of polymorphic bands was % . Conclusion: The 4 samples of Anoectochilus had large ic difference,and the new forms of A. roxburghii were produced probably . The 4 dry materials of Anoectochilus could be distincted by the distinctive bands .

金线莲属兰科(Orchidaceae)开唇兰属(Anoectochilus)植物,该属植物有40多种,分布于亚洲热带地区到大洋洲。我国已经发现的野生种有20种,2个变种,分布于浙江、福建、江西、广东、广西、海南、贵州、云南、西藏、台湾等省区[1]。开唇兰植物具有药用和观赏的功用,但其主要价值在药用。目前,开唇兰植物主要有两个种为药用,即花叶开唇兰(A. roxburghii)和台湾开唇兰(A
