摘要图/表参考文献(27)相关文章(15) 摘要 目的 探索萱草花的抗焦虑作用及其机制。 方法 将60只ICR雄性小鼠采用随机数字表法分成6组,分别为萱草花低剂量组(1 g/kg)、中剂量组(4 g/kg)、高剂量组(8 g/kg)、空白组(生理盐水)、模型组、地西泮阳性对照组(2 mg/kg),每组10只。除空白组外,其余各组均采用慢性束缚法制备小鼠焦虑模型。各组小鼠进行高架十字迷宫、旷场和明暗箱实验。采用酶联免疫吸附试验法测定小鼠脑组织中多巴胺(DA)、5-羟色胺(5-HT)、γ-氨基丁酸(GABA)和谷氨酸(Glu)水平。 结果 与空白组比较,模型组进入中央区的次数减少(P<0.05),中央区停留时间缩短(P<0.01),开臂时间百分比(OT%)和开臂次数百分比(OE%)均降低(P<0.05);进入明箱次数减少,明箱活动时间缩短(P<0.05)。与模型组比较,中、高剂量组进入中央区次数增多(P<0.01);低、中剂量组中央区停留时间延长(P<0.05);低、中、高剂量组OT%均升高(P<0.05或P<0.01);中、高剂量组OE%均升高(P<0.05或P<0.01);中剂量组小鼠进入明箱次数增多(P<0.05);中、高剂量组小鼠明箱活动时间延长(P<0.01)。与空白组比较,模型组小鼠脑组织中GABA含量显著降低,GLU、5-HT、DA含量显著增高(P<0.05)。与模型组比较,低、中、高剂量组小鼠脑中GABA含量均升高(P<0.05或P<0.01);中剂量组小鼠脑中5-HT和DA降低(P<0.05);而高剂量组小鼠脑中GLU、DA含量降低(P<0.05)。 结论 萱草花具有一定的抗焦虑作用,其作用机制可能与增加脑组织 GABA 的水平和降低脑内DA、5-HT的水平有关。服务把本文推荐给朋友加入我的书架加入引用管理器 E-mail AlertRSS作者相关文章王少男1 张韵琦1 景松松1 郑玉光2 孙志勇3关键词 :萱草花, 抗焦虑, 神经递质, 作用机制 Abstract:Objective To determine the anti-anxiety effect of Hemerocallis citrina Borani(XCH) and its mechanism. Methods Sixty male ICR mice were divided into 6 groups by random number table method, including hemerocallis low-dose group (1 g/kg), medium-dose group (4 g/kg), high-dose group (8 g/kg), blank group (normal saline), model group and diazepam positive control group (2 mg/kg), with 10 mice in each group. Except the blank group, the other groups were prepared by chronic restraint stress. Mice of each group underwent elevated cross maze, open field and light and dark box experiments. The levels of dopamine (DA), 5-hydroxytryptamine (5-HT), gamma-aminobutyric acid (GABA) and glutamic acid (Glu) in the brain of mice were determined by enzyme-linked immunosorbent assay. Results Compared with blank group, the number of times of entering the central area decreased (P<0.05), the time of staying in the central area shortened (P<0.01), and the percentage of open arm time (OT%) and the percentage of open arm times (OE%) decreased in model group (P<0.05); the times of entering the open box were reduced, and the activity time of the open box was shortened (P<0.05). Compared with model group, the number of times of entering the central region in medium and high dose groups increased (P<0.01); the residence time in the central region of low and medium dose groups was prolonged (P<0.05). OT% of low, medium and high dose groups was increased (P<0.05 or P<0.01); the OE% of medium and high dose groups was increased (P<0.05 or P<0.01); the number of mice entering the open chamber increased in the medium dose group (P<0.05); the open chamber activity time of mice in medium and high dose groups was prolonged (P<0.01). Compared with blank group, the content of GABA in brain tissue of model group was significantly decreased, while the contents of GLU, 5-HT and DA were significantly increased (P<0.05). Compared with the model group, the GABA level in the brain tissues of the low, medium and high dose groups was increased (P<0.05 or P<0.01). The 5-HT and DA level in the brain tissues of the medium dose group was reduced (P<0.05); the GLU and DA level in the brain tissues of the high dose group was decreased (P<0.05). Conclusion XCH shows obvious anti-anxiety effects, which mechanism may be related to increasing the level of GABA and decreasing the level of DA and 5-HT in the brain tissues.

Compared with the model group, the GABA level in the brain tissues of the low, medium and high dose groups was increased (P<0.05 or P<0.01). The 5-HT and DA level in the brain tissues of the medium dose group was reduced (P<0.05); the GLU and DA level in the brain tissues of the high dose group was decreased (P<0.05). Conclusion XCH shows obvious anti-anxiety effects, which mechanism may be related to increasing the level of GABA and decreasing the level of DA and 5-HT in the brain tissues.
